Among the most important users of plant genetic resources, conserved predominantly in public genebanks around the world, are public and private plant breeders. Through their breeding efforts, they contribute significantly to global, regional, and local food and nutrition security. Plant breeders need genetic diversity to be able to develop competitive new varieties that are adapted to the changing environmental conditions and suit the needs of consumers. To ensure continued and timely access to the genetic resources that contain the required characteristics and traits, plant breeders established working collections with breeding materials and germplasm for the crops they were breeding. However, with the changing and increasingly more restrictive access conditions, triggered by new global legal instruments like the Convention on Biological Diversity/Nagoya Protocol and the International Treaty, plant breeders started to establish their own genebanks at the turn of the 21st century. This paper analyses the conditions that contributed to this situation as well as the historical ways that plant breeders used to acquire the germplasm they needed. Public genebanks played and continue to play a conducive role in providing genetic resources to users, including private-sector plant breeders. However, also the practices of the germplasm curators to collect and distribute germplasm were affected by the new legal framework that had been developed in global fora. It is against this background that the complementarity and collaboration between public and private sector genebanks have been assessed. Whenever possible, vegetable genetic resources and vegetable private breeding companies have been used to analyze and illustrate such collaboration. The authors look at reported successful examples of collaborative efforts and consider opportunities and approaches under which such collaboration can be established and strengthened to ensure the continued availability of the building blocks for food and nutrition security. The vegetation ecosystem in the southern hilly region of China (SHRC) plays a crucial role in the country’s carbon reservoir. Clarifying the dynamics of net primary productivity (NPP) in this area and its response to climate factors in the context of climate change is important for national forest ecology, management, and carbon neutrality efforts. This study, based on remote sensing and meteorological data spanning the period 2001 to 2021, aims to unveil the spatiotemporal patterns of vegetation productivity and climate factors in the southern hilly region, explore interannual variation characteristics of vegetation productivity with altitude, and investigate the response characteristics of NPP to various climate factors. The results indicate that from 2001 to 2021, the annual average NPP in the southern hilly region had a significant increasing trend of 2.13 ± 0.78 g m⁻² a⁻¹. The trend of NPP varies significantly with altitude. Despite a general substantial upward trend in vegetation NPP, regions at lower elevations exhibit a faster rate of increase, suggesting a diminishing difference in the NPP of different elevation ranges. The overall rise in average temperature has positive implications for the southern hilly region, while the impact of precipitation on vegetation NPP demonstrates noticeable spatial heterogeneity. Regions in which vegetation NPP is significantly negatively correlated with precipitation are mainly concentrated in the southern areas of Guangdong, Fujian, and Jiangxi provinces. In contrast, other regions further away from the southeastern coast tend to exhibit a positive correlation. Over the past two decades, there has been an asymmetry in the diurnal temperature variation in the SHRC, with the nighttime warming rate being 1.8 times that of the daytime warming rate. The positive impact of daytime warming on NPP of vegetation is more pronounced than the impact of nighttime temperature changes. Understanding the spatiotemporal patterns of NPP in the SHRC and the characteristics of its response to climate factors contributes to enhancing our ability to protect and manage vegetation resources amidst the challenges of global climate change. BdGF14a, a 14-3-3 gene from Brachypodium distachyon, induced by salt, H₂O₂, and abscisic acid (ABA), improved tolerance to drought and salt in tobacco, with a higher survival rate and longer roots under these stresses. Additionally, physiological index analyses showed that the heterologous expression of BdGF14a induced higher expression levels of antioxidant enzymes and their activities, leading to lighter DAB and NBT staining, denoting decreased H₂O₂ content. Additionally, the lower MDA content and ion leakage indicated enhanced cell membrane stability. Moreover, exogenous ABA resulted in shorter roots and a lower stomatal aperture in BdGF14a transgenic plants. BdGF14a interacted with NtABF2 and regulated the expression of stress-related genes. However, adding an ABA biosynthesis inhibitor suppressed most of these changes. Furthermore, similar salt and drought resistance phenotypes and physiological indicators were characterized in tobacco plants expressing BdbZIP62, an ABRE/ABF that interacts with BdGF14a. And Y1H and LUC assays showed that BdGF14a could enhance the transcription regulation activity of NtABF2 and BdbZIP62, targeting NtNECD1 by binding to the ABRE cis-element. Thus, BdGF14a confers resistance to drought and salinity through interaction with BdbZIP62 and enhances its transcriptional regulation activity via an ABA-mediated signaling pathway. Therefore, this work offers novel target genes for breeding salt- and drought-tolerant plants. Full article

The absence of an externally-imposed 24 h light/dark cycle in closed plant production systems allows setting the light environmental parameters in unconventional ways. Innovative lighting modes for energy-saving, high-quality, and yield production are widely discussed. This study aimed to evaluate the effects of the light/dark cycles of 16/8 h (control) and 24/12 h, 48/24 h, 96/48 h, 120/60 h (unconventional cycles) based on the same total light amount, and continuous lighting (360/0 h) on plant performance of some Solanaceae species. Responses of eggplant (Solanum melongena L.), sweet pepper (Capsicum annuum L.), tobacco (Nicotiana tabacum L.), and tomato (Solanum lycopersicum L.) plants to extended light/dark cycles and continuous lighting were studied under controlled climate conditions. Plants with two true leaves were exposed to different light/dark cycles for 15 days. Light intensity was 250 µmol m⁻² s⁻¹ PPFD, provided by light-emitting diodes (LEDs). After the experiment, tomato, sweet pepper, and eggplant transplants were planted in a greenhouse and grown under identical conditions of natural photoperiod for the estimation of the after-effect of light treatments on fruit yield. Extended light/dark cycles of 24/12 h, 48/24 h, 96/48 h, 120/60 h, and 360/0 h affected growth, development, photosynthetic pigment content, anthocyanin and flavonoid content, and redox state of plants. Effects varied with plant species and length of light/dark cycles. In some cases, measured parameters improved with increasing light/dark periods despite the same total sum of illumination received by plants. Treatments of tomato and pepper transplants with 48/24 h, 96/48 h, and 120/60 h resulted in higher fruit yield compared to conventional 16/8 h photoperiod. The conclusion was made that extended light/dark cycles can result in increased light use efficiency compared to conventional photoperiod and, therefore, reduced product cost, but for practical application, the effects need to be further explored for individual plant species or even cultivars. The most widely used isoprene emission algorithm, G93 formula, estimates instantaneous leaf-level isoprene emission using the basal emission factor and light and temperature dependency parameters. The G93 parameters have been suggested to show variation depending on past weather conditions, but no study has closely examined the relationship between past meteorological data and the algorithm parameters. Here, to examine the influence of the past weather on these parameters, we monitored weather conditions, G93 parameters, isoprene synthase transcripts and protein levels, and MEP pathway metabolites in the tropical tree Ficus septica for 12 days and analyzed their relationship with cumulative temperature and light intensity. Plants were illuminated with varying (ascending and descending) light regimes, and our previously developed Ping-Pong optimization method was used to parameterize G93. The cumulative temperature of the past 5 and 7 days positively correlated with C_T2 and α, respectively, while the cumulative light intensity of the past 10 days showed the highest negative correlation with α. Concentrations of MEP pathway metabolites and IspS gene expression increased with increasing cumulative temperature. At best, the cumulative temperature of the past 2 days positively correlated with the MEP pathway metabolites and IspS gene expression, while these factors showed a biphasic positive and negative correlation with cumulative light intensity. Optimized G93 captured well the temperature and light dependency of isoprene emission at the beginning of the experiment; however, its performance significantly decreased for the latter stages of the experimental duration, especially for the descending phase. This was successfully improved through separate optimization of the ascending and descending phases, emphasizing the importance of the optimization of formula parameters and model improvement. These results have important implications for the improvement of isoprene emission algorithms, particularly under the predicted increase in future global temperatures. Full article

Studies have determined the separate effects of biochar (BC) and manure application on forage species and soil, but few examined the effects of BCs made from different feedstock applied along with dairy manure. We compared the effect of wood- and manure-derived feedstock BC as well as dairy manure amendment application on Cynodon dactylon performance and soil properties in sandy loam and clay loam soils in a greenhouse pot study. Plant samples were assayed for herbage and root dry weight as well as herbage and root N and C percent and yield. Soil samples were assayed for macronutrients, micronutrients, metals, pH and conductivity. Data analyses involved variance analysis and Tukey’s tests using R in RStudio (the IDE). In general, C. dactylon yields or mineral content were not affected by either manure or BC. However, an increase in the total herbage dry weight (30%) and in herbage N% (55%) was observed for clay loam and sandy loam soil, respectively, due to manure amendment application. There were no alterations in clay loam NO₃-N and P due to any treatment; however, in sandy loam, these nutrients were not altered only when wood BC was applied. In sandy loam soil, NO₃-N and P increased when manure BC along with dairy manure and when manure BC alone were applied, respectively. Thus, wood BC application should be considered to avoid these nutrient buildups when dairy manure is used as a soil amendment. This research shows a neutral (BC) or positive (dairy manure amendment) impact on C. dactylon performance. BC incorporation increases soil total C, showing potential for C sequestration. Long-term field trials could corroborate plant performance and soil parameters. Full article

Device structure, light source height, and climatic factors can potentially affect the catching of target pests in light traps. In this study, the installation of an anti-escape cover in a newly designed light trap significantly increased the number of catches of tea leafhoppers, Empoasca onukii, an economically significant pest of tea gardens, and it prevented 97.95% of leafhoppers from escaping. A series of assessments were performed in the field and showed that the optimal trapping window of the light trap was between 1.5 and 2.5 h (2 ± 0.35 h) after sunset, and the starting time of the window was positively correlated with the sunset time. The number of leafhopper catches decreased sharply when the height of the light source was above the flight height range of E. onukii adults. The height of the light source was optimal between 20 and 40 cm above the tea canopy. The efficacy of the light traps for capturing leafhoppers decreased in the autumn peak period. High numbers of leafhopper catches by the newly designed light trap in the summer could reduce E. onukii population sizes in the autumn. Overall, the newly designed light trap can be used to reduce E. onukii adult populations in tea gardens. Full article

In recent years, quinoa, as a nutritious and sustainable food material, has gained increasing popularity worldwide. To investigate the diversity of nutritional characteristics among different quinoa cultivars and explore their potential health benefits, metabolites of five quinoa cultivars (QL-1, SJ-1, SJ-2, KL-1 and KL-2) were compared by non-targeted metabolomics analysis based on UPLC-ZenoTOF-MS/MS in this study. A total of 248 metabolites across 13 categories were identified. Although the metabolite compositions were generally similar among the different quinoa cultivars, significant variations existed in their respective metabolite contents. Among the identified metabolites, amino acids/peptides, nucleosides, saponins and phenolic acids were the most abundant. Notably, SJ-1 exhibited the most distinct metabolite profile when compared to the other cultivars. Amino acids/peptides and nucleosides were found to be crucial factors contributing to the unique metabolite profile of SJ-1. Collectively, these aforementioned metabolites accounted for a substantial 60% of the total metabolites observed in each quinoa variety. Additionally, a correlation between the DPPH radical scavenging activity and the free phenolic content of quinoa was observed. Variations in phenolic content resulted in different antioxidant capacities among the quinoa cultivars, and SJ-1 exhibited lower phenolic levels and weaker antioxidant activity than the others. These results can provide important information for the development of quinoa resources. Full article

A better understanding of the factors that reduce bundle-sheath cell leakage to CO₂ (Փ), enhance 13C carbon isotope discrimination, and enhance the photosynthetic capacity of barley leaves will be useful to develop a nutrient- and water-saving strategy for dry-land farming systems. Therefore, barley plants were exposed to a novel nitrification inhibitor (NI) (3,4-dimethyl-1H-pyrazol-1-yl succinic acid) (DMPSA) and a urease inhibitor (UI) (N-butyl thiophosphorictriamide (NBPT)) with mulched drip fertigation treatments, which included HF (high-drip fertigation (370 mm) under a ridge furrow system), MF (75% of HF, moderate-drip fertigation under a ridge furrow system), LF (50% of HF, low-drip fertigation under a ridge furrow system), and TP (traditional planting with no inhibitors or drip fertigation strategies). The results indicated that the nitrification inhibitor combined with mulched drip fertigation significantly reduced bundle-sheath cell leakage to CO₂ (Փ) as a result of increased soil water content; this was demonstrated by the light and CO₂ response curves of the photosynthesis capacity (An), the apparent quantum efficiency (α), and the ¹³C-photosynthate distribution. In the inhibitor-based strategy, the use of the urease and nitrification inhibitors reduced Փ by 35% and 39% compared with TP. In the NI-HF strategy, it was found that barley could retain the maximum photosynthesis capacity by increasing the leaf area index (LAI), An, rubisco content, soluble protein, dry matter per plant, and productivity. The CO₂ and light response curves were considerably improved in the NI-HF and NI-MF treatments due to a higher 13C carbon isotope (Δ‰), respiration rate (Rd), and Ci/Ca, therefore obtaining the minimum Փ value. With both inhibitors, there was a significant difference between HF and LF drip fertigation. The NI-MF treatment significantly increased the grain yield, total chlorophyll content, WUE, and NUE by 52%, 47%, 57%, and 45%, respectively. Collectively, the results suggest that the new nitrification inhibitor (DMPSA) with HF or MF mulched drip fertigation could be promoted in semi-arid regions in order to mitigate bundle-sheath cell leakage to CO₂ (Փ), without negatively affecting barley production and leading to the nutrient and water use efficiency of barley. Head smut is a soil-borne fungal disease caused by Sporisorium reilianum that infects maize tassels and ears. This disease poses a tremendous threat to global maize production. A previous study found markedly different and stably heritable tassel symptoms in some maize inbred lines with Sipingtou blood after infection with S. reilianum. In the present study, 55 maize inbred lines with Sipingtou blood were inoculated with S. reilianum and classified into three tassel symptom types (A, B, and C). Three maize inbred lines representing these classes (Huangzao4, Jing7, and Chang7-2, respectively) were used as test materials to investigate the physiological mechanisms of tassel formation in infected plants. Changes in enzyme activity, hormone content, and protein expression were analyzed in all three lines after infection and in control plants. The activities of peroxidase (POD), superoxide dismutase (SOD), and phenylalanine-ammonia-lyase (PAL) were increased in the three typical inbred lines after inoculation. POD and SOD activities showed similar trends between lines, with the increase percentage peaking at the V12 stage (POD: 57.06%, 63.19%, and 70.28% increases in Huangzao4, Jing7, and Chang7-2, respectively; SOD: 27.01%, 29.62%, and 47.07% in Huangzao4, Jing7, and Chang7-2, respectively. These were all higher than in the disease-resistant inbred line Mo17 at the same growth stage); this stage was found to be key in tassel symptom formation. Levels of gibberellic acid (GA₃), indole-3-acetic acid (IAA), and abscisic acid (ABA) were also altered in the three typical maize inbred lines after inoculation, with changes in GA₃ and IAA contents tightly correlated with tassel symptoms after S. reilianum infection. The differentially expressed proteins A5H8G4, P09233, and Q8VXG7 were associated with changes in enzyme activity, whereas P49353, P13689, and P10979 were associated with changes in hormone contents. Fungal infection caused reactive oxygen species (ROS) and nitric oxide (NO) bursts in the three typical inbred lines. This ROS accumulation caused biofilm disruption and altered host signaling pathways, whereas NO signaling triggered strong secondary metabolic responses in the host and altered the activities of defense-related enzymes. These factors together resulted in the formation of varying tassel symptoms. Thus, interactions between S. reilianum and susceptible maize materials were influenced by a variety of signals, enzymes, hormones, and metabolic cycles, encompassing a very complex regulatory network. This study preliminarily identified the physiological mechanisms leading to differences in tassel symptoms, deepening our understanding of S. reilianum-maize interactions. Full article

Tryptophan, as a signal molecule, mediates many biotic and environmental stress-induced physiological responses in plants. Therefore, an experiment was conducted to evaluate the effect of tryptophan seed treatment in response to cadmium stress (0, 0.15, and 0.25 mM) in sunflower plants. Different growth and biochemical parameters were determined to compare the efficiency of the treatment agent. The results showed that cadmium stress reduced the growth attributes, including root and shoot length, dry and fresh weight, rate of seed germination, and the number of leaves. Cadmium stress also significantly reduced the contents of chlorophyll a, b, and total chlorophyll, carotenoid contents, phenolics, flavonoids, anthocyanin, and ascorbic acid. Whereas cadmium stress (0.15 and 0.25 mM) enhanced the concentrations of malondialdehyde (45.24% and 53.06%), hydrogen peroxide (−11.07% and 5.86%), and soluble sugars (28.05% and 50.34%) compared to the control. Tryptophan treatment decreased the effect of Cd stress by minimizing lipid peroxidation. Seed treatment with tryptophan under cadmium stress improved the root (19.40%) and shoot length (38.14%), root (41.90%) and shoot fresh weight (13.58%), seed germination ability (13.79%), average leaf area (24.07%), chlorophyll b (51.35%), total chlorophyll (20.04%), carotenoids (43.37%), total phenolic (1.47%), flavonoids (19.02%), anthocyanin (26.57%), ascorbic acid (4%), and total soluble proteins (12.32%) compared with control conditions. Overall, the tryptophan seed treatment showed positive effects on sunflower plants’ growth and stress tolerance, highlighting its potential as a sustainable approach to improve crop performance. Tobacco (Nicotiana tabacum L.) is a major industrial crop that has being cultivated for centuries for the manufacturing of cigarettes, cigars, and other smoking products. Due to its negative effects on both human health and the environment, the European Union has adopted strict policies that aspire to reduce the consumption of tobacco. Herbal cigarettes are alternative smoking products that are often advertised as healthier than conventional tobacco cigarettes and are especially popular in Asian markets. Even though the available literature suggests that they are equally detrimental to human health, the introduction of tobacco-alternative crops (TACs) to the European tobacco industry could smoothen the abandonment of tobacco, and eventually smoking products altogether, in the EU. The aim of the present systematic review was to compile a list of possible TACs that could be incorporated in the European smoking industry, and highlight their strengths and weaknesses. The most dominant crops in the literature (and in the existing market products) were calendula (Calendula officinalis L.), mullein (Verbascum thapsus L.), ginseng (Panax ginseng C.A.Mey.), tea (Camellia sinensis (L.) Kuntze), chamomile (Matricaria chamomilla L.), and mentha (Mentha spp.). Even though these crops are promising, further research is required for their incorporation in the European tobacco industry. Water availability is a limiting factor for the cultivation of sour passion fruit. Soil management techniques and the use of water-retaining polymers can increase soil water retention, reducing the frequency of irrigation in the crop. In this context, the objective of the research was to evaluate the gas exchange, the chlorophyll index, and the yield of the sour passion fruit cv. BRS GA1 as a function of irrigation depths, pit volumes, and doses of water-retaining polymer. The experiment was carried out in randomized blocks, in plots subdivided in a 2 × (2 × 5) arrangement, with irrigation depths of 70 and 100% of the crop evapotranspiration (ETc) as the main plot, the subplots with the volumes of pit of 64 and 128 dm³, and doses of the water-retaining polymer of 0, 0.5, 1.0, 1.5, and 2.0 g dm⁻³. The interaction of irrigation depths × pit volumes × doses of water-retaining polymer influences chlorophyll indexes, gas exchange, and water productivity, with positive impacts on yield of the sour passion fruit. The water depth of 70% of ETc increased the yield of sour passion fruit, in pits of 64 dm³. The application of doses of up to 1.1 g dm⁻³ of the water-retaining polymer and irrigation with water of 70% of ETc is recommended, and a dose of 2.0 g dm⁻³ of the water-retaining polymer in a pit volume of 128 dm³, associated with an irrigation depth of 100% ETc causes stress in sour passion fruit plants due to excess water. Sorghum mosaic virus (SrMV) is one of the most prevalent viruses deteriorating sugarcane production. Salicylic acid (SA) plays an essential role in the defense mechanism of plants and its exogenous application has been observed to induce the resistance against biotic and abiotic stressors. In this study, we set out to investigate the mechanism by which sorghum mosaic virus (SrMV) infected sugarcane responds to SA treatment in two sugarcane cultivars, i.e., ROC22 and Xuezhe. Notably, significantly low viral populations were observed at different time points (except for 28 d in ROC22) in response to post-SA application in both cultivars as compared to control based on qPCR data. Furthermore, the lowest number of population size in Xuezhe (20 copies/µL) and ROC22 (95 copies/µL) was observed in response to 1 mM exogenous SA application. A total of 2999 DEGs were identified, of which 731 and 2268 DEGs were up- and down-regulated, respectively. Moreover, a total of 806 DEGs were annotated to GO enrichment categories: 348 biological processes, 280 molecular functions, and 178 cellular components. GO functional categorization revealed that DEGs were mainly enriched in metabolic processes, extracellular regions, and glucosyltransferase activity, while KEGG annotation revealed that DEGs were mainly concentrated in phenylpropanoid biosynthesis and plant-pathogen interaction suggesting the involvement of these pathways in SA-induced disease resistance of sugarcane in response to SrMV infection. The RNA-seq dataset and qRT-PCR assay showed that the transcript levels of PR1a, PR1b, PR1c, NPR1a, NPR1b, PAL, ICS, and ABA were significantly up-regulated in response to SA treatment under SrMV infection, indicating their positive involvement in stress endorsement. Overall, this research characterized sugarcane transcriptome during SrMV infection and shed light on further interaction of plant-pathogen under exogenous application of SA treatment. Full article

Poplar (Populus spp.) is a valuable tree species with multiple applications in afforestation. However, its growth in saline areas, including coastal regions, is limited. This study aimed to investigate the physiological mechanisms of arbuscular mycorrhizal fungi (AMF) symbiosis with 84K (P. alba × P. tremula var. glandulosa) poplar under salt stress. We conducted pot experiments using NaCl solutions of 0 mM (control), 100 mM (moderate stress), and 200 mM (severe stress) and evaluated the colonization of AMF and various physiological parameters of plants, including photosynthesis, biomass, antioxidant enzyme activity, nutrients, and ion concentration. Partial least squares path modeling (PLS-PM) was employed to elucidate how AMF can improve salt tolerance in poplar. The results demonstrated that AMF successfully colonized the roots of plants under salt stress, effectively alleviated water loss by increasing the transpiration rate, and significantly enhanced the biomass of poplar seedlings. Mycorrhiza reduced proline and malondialdehyde accumulation while enhancing the activity of antioxidant enzymes, thus improving plasma membrane stability. Additionally, AMF mitigated Na⁺ accumulation in plants, contributing to the maintenance of a favorable ion balance. These findings highlight the effectiveness of using suitable AMF to improve conditions for economically significant tree species in salt-affected areas, thereby promoting their utilization. In this study, in vitro antioxidant, antimicrobial, cytotoxic, and cell migration effects of phenolic compounds of Lathyrus tuberosus leaves, known in the Transylvanian ethnomedicine, were investigated. Ultra-high-performance liquid chromatography-tandem mass spectrometry was employed for the analysis of the ethanolic and aqueous extracts. The antimicrobial properties were determined using a conventional microdilution technique. Total antioxidant capacity techniques were used using cell-free methods and cell-based investigations. Cytotoxic effects were conducted on 3T3 mouse fibroblasts and HaCaT human keratinocytes using a multiparametric method, assessing intracellular ATP, total nucleic acid, and protein levels. Cell migration was visualized by phase-contrast microscopy, employing conventional culture inserts to make cell-free areas. Together, 93 polyphenolic and monoterpenoid compounds were characterized, including flavonoid glycosides, lignans, hydroxycinnamic acid, and hydroxybenzoic acid derivatives, as well as iridoids and secoiridoids. The ethanolic extract showed high antioxidant capacity and strong antimicrobial activity against Bacillus subtilis (MIC₈₀ value: 354.37 ± 4.58 µg/mL) and Streptococcus pyogenes (MIC₈₀ value: 488.89 ± 4.75 µg/mL). The abundance of phenolic compounds and the results of biological tests indicate the potential for L. tuberosus to serve as reservoirs of bioactive compounds and to be used in the development of novel nutraceuticals. Oxidosqualene cyclases (OSCs) are the key enzymes accountable for the cyclization of 2,3-oxidosqualene to varied triterpenoids and phytosterols. Hoodia gordonii (from the family Apocynaceae), a native of the Kalahari deserts of South Africa, Namibia, and Botswana, is being sold as a prevalent herbal supplement for weight loss. The appetite suppressant properties are attributed to P57AS3, an oxypregnane steroidal glycoside. At the molecular level, the enzymes involved in the biosynthesis of triterpenes and phytosterols from H. gordonii have not been previously reported. In the current study, predicted transcripts potentially encoding oxidosqualene cyclases were recognized first by searching publicly available H. gordonii RNA-seq datasets. Two OSC-like sequences were selected for functional analysis. A monofunctional OSC, designated HgOSC1 which encodes lupeol synthase, and HgOSC2, a multifunctional cycloartenol synthase forming cycloartenol and other products, were observed through recombinant enzyme studies. These studies revealed that distinct OSCs exist for triterpene formation in H. gordonii and provided opportunities for the metabolic engineering of specific precursors in producing phytosterols in this plant species. Insect larval frass has been proposed as a fertilizer and amendment, but methods for testing its effects on plants are poorly developed and need standardization. We obtained different types of black soldier fly (Hermetia illucens) frass via the factorial combination of (a) two insect diets, as follows: G (Gainesville = 50% wheat bran, 30% alfalfa meal, 20% maize meal) and W (43% sheep whey + 57% seeds); (b) two frass thermal treatments: NT = untreated and T = treated at 70 °C for 1 h. We tested the effects on the germination of cress (Lepidium sativum L.) and wheat (Triticum durum Desf.) by applying 1:2 w:w water extracts at 0, 25, 50, 75 and 100% concentration. Standardizing frass water content before extraction affected chemical composition. Frass extracts showed high electrical conductivity (8.88 to 13.78 mS cm⁻¹). The W diet was suppressive towards Escherichia coli and showed a lower content of nitrates (e.g., WNT 40% lower than GNT) and a concentration-dependent phytotoxic effect on germinating plants. At 25% concentration, germination indices of G were 4.5 to 40-fold those at 100%. Root and shoot length and root hair area were affected by diet and concentration of frass extracts (e.g., root and shoot length in cress at 25% were, respectively, 4.53 and 2 times higher than at 100%), whereas the effects of the thermal treatment were few or inconclusive. On barley (Hordeum vulgare L.) grown in micropots on a silty loam soil, root mass was reduced by 37% at high extract concentration. A quick procedure for root hair surface area was developed based on the geographic information system (GIS) and may provide a fast method for incorporating root hair phenotyping in frass evaluation. The results indicate that below-ground structures need to be addressed in research on frass effects. For this, phyotoxicity tests should encompass different extract dilutions, and frass water content should be standardized before extraction in the direction of canonical procedures to allow comparisons. The agro-pastoral ecotone in northern China is the main production area of agriculture and animal husbandry, in which agricultural development relies entirely on groundwater. Due to the increasing water consumption of groundwater year by year, groundwater resources are becoming increasingly scarce. The substantial water demand and low germination rate in the first year are the main characteristics of alfalfa (Medicago sativa L.) yield in the agro-pastoral ecotone in northern China. Due to unscientific irrigation, water resources are seriously wasted, which restricts the development of local agriculture and animal husbandry. The study constructed the Dssat-Forages-Alfalfa model and used soil water content, leaf area index, and yield data collected with in situ observation experiments in 2022 and 2023 to calibrate and validate the parameters. The study found ARE < 10%, E_NRMS < 15%, and R² ≥ 0.85. The model simulation accuracy was acceptable. The study revealed that the water consumption at the surface soil layer (0–20 cm) was more than 6~12% and 13~31% than that at the 20–40 cm and 40–60 cm soil layers, respectively. The study showed when the irrigation quota was 30 mm, the annual yield of alfalfa (Medicago sativa L.) (7435 kg/ha) was consistent with that of the irrigation quota of 33 mm, and increased by 3.99% to 5.34% and 6.86% to 10.67% compared with that of irrigation quotas of 27 mm and 24 mm, respectively. To ensure the germination rate of alfalfa (Medicago sativa L.), it is recommended to control the initial soil water content at 0.8 θ_fc~1.0 θ_fc, with an irrigation quota of 30 mm, which was the best scheme for water-use efficiency and economic yield. The study aimed to provide technological support for the rational utilization of groundwater and the scientific improvement of alfalfa yield in the agro-pastoral ecotone in northern China.
